What Is the Endocannabinoid System?

The endocannabinoid system, or ECS, is a signalling network made of endocannabinoids produced by the body, receptors including CB1 and CB2, and enzymes that create and break down those molecules. It helps regulate signalling related to functions such as appetite, memory, pain, movement, stress and immune activity. Cannabis compounds can influence this system.

What are endocannabinoids?

Endocannabinoids are lipid-based signalling molecules made by the body. Two of the best studied are anandamide, also called AEA, and 2-arachidonoylglycerol, or 2-AG. They are produced when needed and are then broken down by enzymes.

They are different from phytocannabinoids, which come from plants. THC and CBD are phytocannabinoids.

What are CB1 and CB2 receptors?

CB1 receptors are abundant in the central nervous system, although they also occur elsewhere. Their distribution helps explain why THC can affect memory, movement, appetite, perception and coordination.

CB2 receptors are strongly associated with immune and peripheral tissues, but the simple “CB1 in the brain, CB2 in the body” slogan is incomplete because both receptor types have broader distributions.

How does THC interact with the ECS?

THC can activate CB1 receptors and produce intoxication and impairment. Its effects are stronger and longer-lasting than the body’s tightly controlled endocannabinoid signals, particularly at larger doses.

How does CBD interact with the ECS?

CBD does not activate CB1 receptors in the same way as THC and does not produce a high. Its pharmacology is complex and involves multiple molecular targets. Claims that CBD simply “balances” or “boosts” the ECS are marketing shorthand, not a complete scientific explanation.

What does the ECS regulate?

Research links ECS signalling with appetite, pain processing, memory, mood, stress responses, movement, reward, sleep and immune functions. That does not mean cannabis improves every process associated with the ECS. Changing a regulatory system can produce both desired and unwanted effects.
